Greetings:
I have the attached leave schedule for staff. Is it possible to highlight the date cells as shown in the excel sheet based on the changes in the data in column B, C & E.

You can do this using Conditional Formatting. To view the rules, select Conditional Formatting > Manage Rules... on the Home tab of the ribbon.

I would adopt a data validation method.

I had managed it by adding additional cells for dates as shown in the attached.
Now I want to highlight the cells where another cell contains a specific value (attached the sample) I had tried but not successful.

Your sample workbook doesn't explain what you want to do...

Select V5:AY13. The active cell in the selection should be V5.
Create a new conditional formatting rule of type formula with formula
=V$3="H"
Here, V is the column of the active cell; this is relative because it must change for the other columns.
3 is the row that may contain "H". This is absolute since it should not change for the other rows.
